5-Hydroxytryptophan (5-HTP) is a metabolite of L-tryptophan and the precursor to the neurotransmitter serotonin. It has superior bioavailability through oral dosing and readily converts into serotonin in the brain and body. This metabolite form bypasses multiple barriers, such as carrier requirements, competitive inhibition, and shunting to other pathways that could inhibit the conversion of tryptophan to serotonin.

By increasing serotonin in the central nervous system, 5-HTP has clinical implications ranging from sleep and mood disorders to fibromyalgia and pain perception.

Mood Disorder

Over the past 40 years a multitude of studies have sought to confirm 5-HTP's effect on depression. Most placebo controlled trials demonstrated positive clinical outcomes, such as reduction in depressive symptoms. Sano initiated one of the larger trials in the early 1970's noting significant improvement in depressive symptoms in the majority of patients (with a 50-100mg dose) and a relatively quick response rate; under two weeks. Subsequent Japanese studies were able to replicate this positive outcome. The majority of subjects reported benefits within two weeks and as little as three days for some. A study comparing 5-HTP to a selective serotonin reuptake inhibitor (SSRI) demonstrated that while both agents had similar beneficial effects on depressive symptoms, 5-HTP had less reported side effects.

Due to its known promotion of serotonin, 5-HTP has historically been used for anxiety-related issues, however, minimal human trials have been conducted. An in vivo study by Carnevale et al. demonstrated a significant anxiolytic effect of Griffonia seed extract (supplemental source of 5-HTP) dosed at 5-25mg/kg.

Sleep Disturbance

Studies evaluating the efficacy of 5-HTP for insomnia have demonstrated significant results. Consuming an oral dose of 200-600mg of 5-HTP before bed has been shown to increase REM sleep, translating into improved quality of sleep. A recent randomized controlled trial evaluated the effectiveness of a GABA/5-HTP preparation concluding it decreased length of time to fall asleep, increased the duration of sleep and improved overall quality.


Fibromylagia (FM) is a multi-symptom syndrome characterized by muscular aching and tenderness, severe fatigue, sleep issues and mood disorder. Studies indicate patients with FM are commonly low in serotonin. Clinical trials of 5-HTP therapy for FM revealed significant benefit with dosing ranging from 300-400mg per day. Findings consistently demonstrated improvement in mood, sleep, pain, tender points and stiffness.

Weight Management

5-HTP can promote weight loss by spontaneously decreasing calorie intake and carbohydrate cravings in obese populations. Trials suggest dosing requirements are high, approximately 900mg daily.


As one of the most abundant cofactors in the body, Vitamin B6 plays a key role in the central nervous system production of neurotransmitters, serotonin and GABA. These neurotransmitters have long been known to modulate mood, especially depression and anxiety, and sleep making these conditions amenable to B6 status. Deficiency has been shown to increase sympathetic output, psychological distress and disturbed sleep patterns. Increasing tissue levels of pyridoxine through high-dose supplementation is thought to have beneficial impact on dysphoric mental states.


Glycine is an amino acid that, in addition to its role in protein structure, acts as an inhibitory neurotransmitter affecting both glutaminergic and glycinergic receptors. Three studies assessed low dose (3g) glycine supplementation prior to sleep. Outcomes revealed improved sleep quality, shortened sleep latency, reduced time to REM sleep and improved following day performance. In conjunction with 5-HTP and B6, this small amount of glycine can further promote the overall calming effect of Cyto-Calm.
